Endotoxin Testing: Why Bacterial Residue Matters in Research Material

Endotoxin is among the most consequential contaminants in research material used for cell culture, and among the least visible. It is not detectable by purity chromatography.
What endotoxin is
Endotoxins are lipopolysaccharides (LPS) from the outer membrane of Gram-negative bacteria. They are heat-stable, survive many sterilisation procedures, and persist after the bacteria themselves are gone. Contamination can enter through water, glassware, buffers, or handling rather than through the peptide synthesis itself.
Why it confounds experiments
LPS is a potent agonist of Toll-like receptor 4 (TLR4)3. In cells expressing TLR4 and its co-receptors, picogram-per-millilitre quantities are sufficient to trigger NF-κB signalling and downstream release of inflammatory cytokines. If a researcher is studying an inflammatory or immune-signalling pathway, endotoxin contamination can produce exactly the readout the peptide is being tested for. The observed effect is then attributable to the contaminant, not the compound.
How it is measured
The standard assay is the Limulus amebocyte lysate (LAL) test1, which exploits the clotting cascade of horseshoe crab hemolymph in response to LPS. Variants include gel-clot, turbidimetric, and chromogenic formats, with results reported in endotoxin units per milligram (EU/mg). Recombinant Factor C assays2 offer a synthetic alternative that avoids animal-derived reagents.
Reading the specification
An endotoxin figure is only interpretable with its units and method. For research material destined for cell-based work, a documented low EU/mg value is a meaningfully stronger specification than purity alone, because it addresses a failure mode purity testing structurally cannot detect.
