Heavy Metals and Residual Solvents: The Rest of the Analytical Panel

Purity and identity dominate discussion of peptide documentation, but a complete analytical panel screens for several classes of process residue that neither test captures.
Residual solvents
Solid-phase peptide synthesis and subsequent cleavage rely on organic solvents and scavengers — commonly dimethylformamide, dichloromethane, acetonitrile, diethyl ether, and trifluoroacetic acid. Traces can persist in the finished lyophilised solid. Residual solvents are typically quantified by gas chromatography and evaluated against established categories that group solvents by toxicological concern, with the most hazardous expected to be absent rather than merely limited.
Heavy metals
Elemental impurities can enter from reagents, catalysts, or equipment contact surfaces. Screening is generally performed by inductively coupled plasma mass spectrometry (ICP-MS), which detects elements at trace concentrations. Copper-containing peptides are a special case: for a compound such as GHK-Cu, copper is a deliberate constituent of the molecule rather than a contaminant, and the specification must distinguish between the two.
Water content
Lyophilised peptides are hygroscopic and readily reabsorb atmospheric moisture. Water content is measured by Karl Fischer titration. The figure matters twice: it affects the accuracy of any mass-based calculation, and elevated moisture accelerates hydrolytic degradation during storage.
Why the full panel matters
Each assay addresses a failure mode invisible to the others. A lot can be chromatographically pure, correctly identified, and still carry solvent residue or moisture sufficient to affect experimental reproducibility. Documentation across the full panel is what makes material genuinely characterised rather than merely tested.
